Pharmacology Griseofulvin is a fungistatic agent that disrupts mitosis in fungal cells.
Active against dermatophytes; ineffective against yeasts or other fungal infections.
Adult Dosage PO (microsize):
- 0.5–1 g/day in a single dose or 2–4 divided doses
PO (ultramicrosize):
- 330–660 mg/day in 1–2 divided doses
Duration:
- General skin infections: ≥3 weeks
- Palms/soles: 4–8 weeks
- Nail infections: 6–12 months
Take with meals to enhance absorption; avoid prolonged sun exposure and alcohol.
Pediatric Dosage PO (microsize):
- 11 mg/kg/day
PO (ultramicrosize):
- 7.3 mg/kg/day
Administer as for adults (single or divided doses).
Dosage Forms Microsize:
- Capsule: 250 mg
- Tablet: 250 mg, 500 mg
- Suspension: 25 mg/mL
Ultramicrosize:
- Tablet: 125 mg, 165 mg, 250 mg, 330 mg
Adverse Reactions Occasional:
- Nausea, vomiting
Rare:
- Photosensitivity reactions, peripheral neuritis, leukopenia
Other:
- Can exacerbate acute intermittent porphyria
